DPP-4 selectively cleaves two amino acids from peptides, such as GLP-1 and GIP, which have proline or alanine in the second position (Figure 2). At the active site where DPP-4 has its effect, there is a characteristic arrangement of three amino acids, Asp-His-Ser. Since alanine and proline are crucial for the biological activity of GPL-1 and GIP, they are inactivated by cleaving away these amino acids. Thus, preventing the degradation of the incretin hormones GLP-1 and GIP by inhibition of DPP-4 has potential as a therapeutic strategy in the treatment of type 2 diabetes.
